Our research interests are centered on understanding the structure and functions of carbohydrates. We prepare the polysaccharides and their derived oligosaccharides from marine organism based on feasible strategies to construct the marine carbohydrate database, and develop novel analytical approaches to decode their fine complex structure using sophisticated NMR and biomass spectrometry combined glycobioinformatics. Based on above structural information and combined computer simulation, carbohydrate microarray, and cell or animal-level bioactivities screening methods, the structure–activity relationships of marine polysaccharides try to be illustrated to discover interesting marine bioactive carbohydrate. Besides, we also devote to resolve the critical pharmacokinetic study of carbohydrate-based drugs, to elucidate the mechanisms of their absorption, metabolism, transport and excretion. These studies will lay the foundation for the research and development of marine carbohydrate-based drugs. Another research direction in our lab focuses on elucidating the structure and physiologicalfunction of endogenous glycans (N-glycan, O-glycan, glycosaminoglycans and glycanlipids) in human. Our research has focused on the development of high-throughput technologies to isolate and identify the glycome related to human health and disease, especially the profile of glycome in the clinical samples. Based on the comprehensively evaluation of the glycome in the different progress of critical illness, we attempt to elucidate the variation of glycans and find the specific glycan. These findings could allow the identification of relevant glycan biomarkers for the diagnosis, early detection, and monitoring of cancer treatment with sufficient sensitivity and specificity for clinical use, which could also enhance understanding of critical illness. |
